Abstract
Cleaned oocysts of Eimeria bovis, E. ellipsoidalis, E. auburnensis, and E. zurnii of cattle, E. ahsata and E. ninakohlyakimovae of sheep, E. bilamellata and E. callospermophili of the ground squirrel, and E. tenella of chickens were pretreated with 50 percent carbon dioxide/50 percent air at temperatures ranging from 29 to 45 C with 2 degree intervals for 4, 6, 8, 10, and 30 hours and treated with bile and trypsin.
The optimum temperature in species of ruminant coccidia was 39 to 41 C. In E. bovis, excystation occurred from 29 to 43 C. At 43 C and above, disintegration of sporozoites was observed within the oocysts. At 29 C, little or no excystation occurred except when the duration of pretreatment was extended to 30 hours. The temperature at which pretreatment occurred was by far more important than that for treatment in the process of excystation.
When all the bovine, ovine, squirrel, and chicken species under study were compared, it was observed that E. ellipsoidalis excysted faster than any of the other species. With 10 hours of pretreatment at 39 C, each of the four bovine species had distinctly different patterns of excystation. The two ovine species, E. ahsata and E. ninakohlyakimovae paralleled E. auburnensis and E. bovis, respectively, in their pattern of excystation. The two species of ground squirrel coccidia did not excyst under any of the conditions in the series of experiments. E. tenellaof this chicken did not respond either, except for oocysts pretreated 30 hours at 39 C. Intact oocysts of E. tenella pretreated with 90 percent carbon dioxide/10 percent air for 24 hours at 39 C excysted at a high level.
