Induction of Hypoxylon cankers in Aspens by artificial inoculation

Abstract

Inoculations using mycelia of Hypoxylon pruinatum (Klot.) Cke. grown on (1) oat-wheat, (2) bran, and (3) Potato-cerelose-agar media were compared with spore inoculations in covered and uncovered wounds on Populus tremuloides Michx. and P. grandidentata Michx. in five experimental plots in northern Wisconsin. Infections occurred and typical Hypoxylon cankers developed following inoculations with mycelia but not with spores. Highly significant differences in percent infection were found among plots and type of inocula but not between presence or absence of covering. A 3:1 oat-wheat medium was significantly superior to other media tested for mycelial inoculations and gave from 70 to 100 percent infection.
