Colchicine treatment of newly formed embryos of Quaking Aspen

Abstract
A technique is presented for treating newly formed embryos of Populus tremuloides Michx. with colchicine to produce tetraploid plants. Pollinated catkins were immersed in a 0.3 percent solution of colchicine for 6 hours, with treatments starting 6, 12, 18, 24, and 30 hours after pollination.The procedure has the advantage that only the relatively few cells of young embryos are involved in the treatment, thereby greatly increasing the chances of obtaining completely tetraploid individuals.
